چکیده

Abstract Oil from Nigella sativa seeds (“black seed oil”) has been used for health care and medical purposes over 2000 years. The major active compound thymoquinone (TQ) shown to have immune regulatory effects in various diseases. Tumor microenvironment is critical tumor development. Immune cells, particularly T are central players kill cells affect progression metastasis. impact of TQ on human cell-mediated anti-tumor response was investigated. Using PBMCs, we activated with anti-CD3/CD28 vitro, treated TQ. First, tested the cell activation, proliferation apoptosis; second, effect after treatment TQ; last, determined alterations gene expression at RNA level treatment. increased activation days 2–4 more activation-induced death day 3. After 7–11 days, TQ-treated showed less survival than control medium. At low concentration (10uM), induced CD3+ proliferation; while 20uM 50uM cytotoxic death. When co-cultured multiple types cancer exhibited significantly enhanced activity, indicated by apoptosis expansion. Gene study upregulation NFAT signaling pathway, including 10+ fold up-regulation IL-2, which might explain In summary, can enhance vitro. This immunoregulatory function could benefit patients different cancers. Supported grants NIH/ NIGMS (2U54GM104942-02, 5U54GM104942-03).

Cyclooxygenase-2 inhibitor treatment enhances photodynamic therapy-mediated tumor response.

Photodynamic therapy (PDT) continues to be used in the treatment of solid tumors. Clinical results are promising, but the therapy has not been optimized, and tumor recurrences can occur. Recently, it has been shown that inhibitors of cyclooxygenase (COX)-2 can be effective in combination with conventional chemotherapy and radiation therapy. In the current study, we examined the parameters of PD...

Tumor sialylation impedes T cell mediated anti-tumor responses while promoting tumor associated-regulatory T cells

The increased presence of sialylated glycans on the tumor surface has been linked to poor prognosis, yet the effects on tumor-specific T cell immunity are hardly studied. We here show that hypersialylation of B16 melanoma substantially influences tumor growth by preventing the formation of effector T cells and facilitating the presence of high regulatory T cell (Treg) frequencies. Knock-down of...

Talilmogene Iaherparepvec generates systemic T-cell-mediated anti-tumor immunity

Talimogene laherparepvec is an investigational oncolytic immunotherapy based on a modified herpes simplex virus type-1 (HSV-1) that is designed to selectively replicate in tumor tissue and to stimulate a systemic antitumor immune response. Intralesional administration of talimogene laherparepvec is intended to result in oncolysis within injected tumors. Iterative viral replication of virus with...

Thymoquinone enhances cisplatin-response through direct tumor effects in a syngeneic mouse model of ovarian cancer

BACKGROUND Ovarian cancer is the most lethal gynecologic malignancy characterized by the frequent development of resistance to platinum chemotherapy. Finding new drug combinations to overcome platinum resistance is a key clinical challenge. Thymoquinone (TQ) is a component of black seed oil that exerts multiple anti-tumorigenic effects on cells, including inhibition of NF-κB and promotion of DN...

Virus-stimulated neutrophils in the tumor microenvironment enhance T cell-mediated anti-tumor immunity

The tumor microenvironment (TME) fosters tumors by attenuating anti-tumor immunity, reinforcing tumor cell survival and increasing angiogenesis. Among the constituents of the TME, here, we focused on tumor-associated neutrophils (TANs). First, we found that the combination of poly I:C and inactivated Sendai virus particles (hemagglutinating virus of Japan envelope; HVJ-E) synergistically suppre...
